Breaking Down the Enigma of Dry Eye

Methionine uptake and metabolism contributes to cancer pathogenesis through functions in methylation reactions and one-carbon metabolism. This Review discusses methionine metabolism in the context of nutrition and the potential of targeting methionine metabolism in cancer through dietary or pharmacological intervention. The interactions between tumours and the immune system are highly complex. This article discusses methods — primarily computational tools — for characterizing diverse aspects of cancer—immune cell interactions, including antigen presentation, T cell repertoires and heterogeneity in cell types and cell states.

The Review particularly highlights the insights from single-cell data from both sequencing technologies and in situ imaging of tissues. This Review discusses the various approaches that are being explored to target transcription factors in cancer, with many of the inhibitors developed from such approaches now advancing to early clinical trials. Schneider et al.

They discuss alterations of these immune regulators, their clinical relevance and therapeutic targeting strategies. Clinical application of CAR T cells to cancer therapy has met with much success, but future trials must build on current understanding to overcome therapy resistance. Review Article 9 Sep Nature Medicine. Previous nanomedicine approaches have attempted to concentrate the action of cytotoxic therapies at tumours, generally with limited success. This Review discusses how the field is evolving to use nanoparticles and biomaterials to program the location, pharmacokinetics and co-delivery of immunotherapies, eliciting responses that cannot be achieved upon administration of such compounds in solution.

In this Review, Sebestyen et al. Metabolic glycoengineering provides a strategy to manipulate glycan structure and function to detect, monitor and treat disease. In this Review, we discuss the chemistry underlying metabolic glycoengineering and highlight recent advances towards the clinical translation of this technology. News 18 Sep Nature. Why brain tumours progress rapidly is unclear. The finding that such cancer cells form synaptic connections with neurons uncovers an interaction that accelerates tumour growth rate and lethality.

The presence and prognostic relevance of the intratumoural microbiota in pancreatic cancer, and the roles of intratumoural bacteria in oncogenesis and therapeutic response are beginning to be elucidated. The feasibility of characterizing intratumoural microbial communities from paraffin-embedded tissues has now been validated, providing greater opportunities for retrospective research. Correspondence 17 Sep Nature. Rivadeneira et al.

Vaccinia -mediated engineered expression of leptin in cancer cells improved responses through metabolic reprogramming of T cells. In this issue, Diehl et al. Cell lines are valuable tools for developing treatments to minimize disparities in prostate cancer outcomes.

Alzheimer's causes chronic, low-level brain cell inflammation. Researchers are studying ways to treat inflammatory processes at work in Alzheimer's disease. The drug sargramostim Leukine is currently in research. It's thought that the drug may stimulate the immune system to protect the brain from harmful proteins. Researchers studied the diabetes drug pioglitazone Actos because it may lessen beta-amyloid and inflammation in the brain, but this trial was negative. Researchers are studying the effects of insulin on the brain and brain cell function, and insulin changes in the brain that may be related to Alzheimer's.

A trial testing an insulin nasal spray to determine whether it slows the progression of Alzheimer's was recently reported as negative. Growing evidence suggests that brain health is closely linked to heart and blood vessel health. The risk of developing Alzheimer's appears to increase as a result of many conditions that damage the heart or arteries. These include high blood pressure, heart disease, stroke, diabetes and high cholesterol. A number of studies are exploring how best to build on this connection.

Strategies under investigation include:.

Alzheimer's treatments: What's on the horizon?

In one study, taking estrogen-based hormone therapy for at least a year during perimenopause or early menopause appeared to protect thinking and memory in women with a higher risk of Alzheimer's disease. But further research has been conflicting, with some studies indicating that estrogen didn't offer any benefit. More research and a better understanding of the relationship between estrogen and cognitive function are needed before any recommendations can be made.

Developing new medications is a slow and painstaking process. The pace can be especially frustrating for people with Alzheimer's and their families who are waiting for new treatment options.

Advisory Council December Meeting Presentation: Research Progress Report | ASPE

To help accelerate discovery, the Coalition Against Major Diseases CAMD , an alliance of pharmaceutical companies, nonprofit foundations and government advisers, has forged a first-of-its-kind partnership to share data from Alzheimer's clinical trials. Researchers anticipate that these data standards and the sharing of data from more than 6, study participants will speed development of more-effective therapies.
